It works by increasing the level of acetylcholine and helps in controlling the symptoms of the disease.

• Ameloss 5 does not cure Alzheimer's disease but may help improve brain function and the ability to do daily activities.
• Take it with or without food, preferably at night before going to bed.
• It may cause diarrhea, nausea, and vomiting. Drink plenty of fluids to keep yourself hydrated while taking Donepezil.
• Monitor your body weight regularly as both Donepezil and  Alzheimer's disease can cause weight loss.
• Do not drive or do anything that requires mental focus until you know how Donepezil affects you.
• Be cautious while taking this medicine if you suffer from peptic ulcer as it may cause vomiting, stomach bleeding, stomach pain, or discomfort.
• Inform your doctor if you develop muscle tremors, especially if you have Parkinson’s disease.
• Inform your doctor if you experience seizures or convulsions while taking this medicine.

Indication of Ameloss 5

Donepezil is used in the treatment of dementia associated with Alzheimer's disease. It is not recommended in patients with gastrointestinal bleeding and lung diseases. 

Pharmacology

Donepezil Hydrochloride is a centrally-acting anticholinesterase agent. It binds reversibly to acetylcholinesterase, inhibiting the hydrolysis of acetylcholine, leading to an increase in acetylcholine concentration at cholinergic synapses in the brain, which may improve cognitive function in Alzheimer's disease.

Dosage & Administration of Ameloss 5

• Initial Dose: 5 mg once daily at bedtime for at least one month to assess clinical response and to achieve steady-state concentrations.
• Dose Adjustment: After one month, the dose may be increased to 10 mg once daily.
• Renal & Hepatic Impairment: No dose adjustment is required for mild to moderate impairment.
• Pediatric Use: Not recommended, as there are no adequate safety and efficacy trials in children.

Take Donepezil at the same time every day. Do not take in larger amounts than advised. Consult the doctor if you experience any undesirable effects. Do not take this medicine if you have any gastrointestinal disease and lung diseases.

Interaction

• Avoid Anticholinergic drugs (e.g., atropine, benztropine) as they may counteract the effects of Donepezil.
• May reduce Donepezil’s effect: Carbamazepine, dexamethasone, phenobarbital, phenytoin.
• May increase Donepezil’s effect: Ketoconazole, quinidine, cimetidine.

Contraindications

Hypersensitivity to Donepezil Hydrochloride or piperidine derivatives.

Pregnancy & Lactation

• Pregnancy: Use only if the potential benefit outweighs the potential risk to the fetus, as there are no adequate studies in pregnant women.
• Lactation: It is unknown if Donepezil is excreted in human breast milk; not recommended for nursing mothers.

Precautions & Warnings

• Use with caution in patients with:
  • Sick sinus syndrome or other heart conduction issues.
  • Risk of peptic ulcers.
  • Asthma or obstructive airway disease.
  • Undergoing anesthesia.

• Keep out of reach of children.
